Simple Sperm Wash is a time tested method for IUI preparation from the beginning of ART. This involves a basic centrifugation step after diluting the semen sample with the media. The dilution step is crucial for breaking the viscosity and allows sedimentation of cells during centrifugation. Repeating the centrifugation step further cleans the sperm from the semen contents. Finally, the sedimented pellet is subjected to swim-up for clean sperm separation.

In the Direct Swim-up method, a fraction of the semen sample is underplayed in the test tube containing 2ml of the media and incubated at 37 C temperature. Sperms directly swim into the media giving a very gentle separation of sperms without any stress.
